The effect of wortmannin on IgG-receptor (FcyR)-mediated
stimulation of human neutrophils was investigated. The Ca^(2+)
influx induced by clustering of both Fcy receptors was inhibited
by wortmannin, as was the release of Ca^(2+) from intracellular
stores. Wortmannin also inhibited, with the same efficacy, the
accumulation of Ins(1,4,5)P3 observed after FcyR stimulation,
but did not affect the increase in Ins(1,4,5)P3 induced by the
chemotactic peptide, formyl-methionine-leucine-phenylalanine.
Because wortmannin is, in the concentrations used here, an
inhibitor of PtdIns 3-kinase, these results suggested a role for
PtdIns 3-kinase upstream of Ca^(2+) signalling, induced by FcyR
cross-linking. Support for this notion was obtained by investigating
the effect of another inhibitor of PtdIns 3-kinase, LY
294002, and by studying the kinetics...
This thesis is on light damage to the retina. Photochemical light damage to the retina can
even occur under ambient light conditions as demonstrated by Noell et al. (1966) They found
green light (500 nm) the most powerful in causing retinal damage. Under higher irradiance
conditions the action spectrum changes: Short wavelengths are then the most harmful (Ham et
al., 1979; Ham, Mueller and Sliney, 1982; Gorgels and van Norren, 1995). With ever
increasing irradiance the domain of thermal damage is entered. The highest retinal irradiance
used in the experiments in this thesis is approximately 300 mW/ cm . This is a rather high level, 2
but insufficient to increase the retinal temperature over 10 C; the generally accepted safety o
limit (Ham et al., 1979). Another argument against thermal damage, is the duration of the
exposure. Thermal d...
De juiste diagnose, classificatie en behandeling van thoraco-lumbale wervelfracturen blijven een problematisch en controversieël
onderwerp in traumatologie.
Abstract. In patients suffering from temporal lobe epilepsy (TLE) a highly variable degree of hippocampal sclerosis (HS) can be observed. For standard neuropathological evaluation after hippocampal resection, neuronal cell loss in the hippocampal subareas is assessed (Wyler score 0-4) [Wyler et al. (1992) J Epilepsy 5: 220-225]. Other marked morphological changes in the sclerotic hippocampus are gliosis and loss of mossy fibers in the hilus and mossy fiber sprouting in the supragranular layer. In this study we quantified changes in mossy fiber density using Timm's stain in resected hippocampal tissue from patients with various degrees of sclerosis. We found that tissue specimens from patients without sclerosis (W0) show almost no mossy fiber sprouting. Patients with moderate sclerosis show sprouting without fiber loss in the hilus, whe...
